A reliable, accurate and sensitive RT-PCR test for detecting Wheat streak mosaic virus (WSMV) in bulk samples of wheat seed was successfully developed, validated in blind studies and made available to the agricultural industry through AGWEST Plant laboratories for use in testing seed stocks... The three main ways to combat WSMV are to 1) destroy the green bridge before sowing to prevent transmission of the virus by the WCM, 2) use wheat seed stocks that have either none or only low levels of the virus, and 3) to sow crops late when temperature conditions are non-conducive for WCM... In an early seed transmission experiment, WSMV was detected by RT-PCR in seedlings nine days after sowing and the number of positive seedlings increased considerably over the following six weeks until almost all batches tested positive...

Use of the test may also help to prevent wheat breeders sowing infected seed stocks... Use of the test confirmed widespread occurrence of WSMV in the WA grainbelt and identified the disease for the first time in Tasmania... The research also showed that Australian WSMV isolates represent a single incursion from overseas and the virus has been present in Australia for at least 15 years...

" Attachment 1 Project DAW00141 Developing a sensitive dry seed test to detect seed-borne viruses in bulk seed samples of wheat Procedure for RT-PCR detection of WSMV in bulk seed samples of wheat [Procedure developed for detection of Wheat streak mosaic virus (WSMV) in dry, bulk seed samples of wheat seed during GRDC project DAW 00141 "Developing a sensitive dry seed test to detect seed-borne viruses in bulk seed samples of wheat"... The test does not provide any quantitative indication of the precise level of WSMV seed transmission to seedlings in the seed stock tested which may be crucial information for growers when seed is to be sown on farms with a previous history of WSMV in their crops... The dry, bulk seed test is based on RT-PCR so its extremely high sensitivity means that great caution must be taken to avoid contamination of seed with dust that may carry wheat micro-trash contaminated with WSMV...
