The SAGIT funded project involves using a controlled environment assay that is run at Roseworthy, to identify and quantify the level of heat stress tolerance in current varieties, advanced breeders lines and exotic lines, as well as using 'gene mapping', to genetically dissect and understand the key sources of tolerance identified to date... Figure 1 illustrates the fertility and grain size response for a subset of varieties to heat stress in the controlled environment assay. In general heat stress causes a significant (p<0.05) reduction in both grain number and grain size, the two key yield components and ultimately grain yield... To address the concerns, we will identify chromosome regions governing variation in heat tolerance and deliver DNA-based genetic markers that breeders can use to select for the favourable chromosome variants... Chromosome regions governing heat tolerance traits will be identified by following segregation in experimental populations derived from bi-parental crosses, as well as by identifying marker-trait associations across a broad collection of elite and exotic germplasm... Lines possessing heat tolerance traits identified in greenhouse/chamber experiments (maintenance of chlorophyll and single grain weight) will also be evaluated in the field to establish the relevance of these assays to heat tolerance breeding...

Barley with genetically unique sources of adaptation to the low rainfall environments has been evaluated using lines from the International Centre for Agricultural Research in the Dry Areas (ICARDA)... Barley lines from ICARDA have shown potential for the improvement of adaptation, with adaptive features derived from wild barley and primitive barley landraces, and selection for yield and associated adaptive traits in drought prone environments with soils of low fertility... The germplasm and information gathered in this project will benefit industry since the project has had a focus on the improvement of adaptation as related to yield and physical grain quality...

The aim of the project was to develop closely linked molecular markers in wheat for traits that were suggested by breeders or collaborative partners at the Molecular Plant Breeding Cooperative Research Centre (MPBCRC)... All three objectives were achieved: two closely linked markers (< 20kb) to the 7A maturity locus were identified and further alleles were suggested to segregate in Australian germplasm; two markers were developed for the 2A rust gene (< 2cM) and marker density at three drought-tolerance related loci was increased by adding 53 markers out of more than 200 screened simple sequence repeats (SSRs)... The genetic map of the cross between Berkut and Krichauff can be seen as a valuable genetic resource indicated by the many projects on QTL identification in this population such as yield under drought stress, salinity in hydroponics and field and grain fructan content...

Unpredictable weather conditions at the time of flowering cause large yield fluctuations on a regular basis... Stabilising productivity of wheat crops in an unpredictable and changing environment requires wheat varieties that are more tolerant to abiotic stress conditions (terminal drought, heat, chilling and frost)... We have identified the genetic loci responsible for maintaining grain number in two wheat doubled haploid populations (Cranbrook x Halberd, Sundor x AUS30604) using hydroponics and osmotic stress as an alternative screening method and are currently evaluating drought tolerance of selected lines of these populations under field conditions...
